草地藏系绵羊心脏脂肪酸结合蛋白基因第二内含子的克隆和测序
Clone and Sequence of the Second Intron of Heart Fatty Acid Binding Protein Gene of Tibetan Sheep in Pasture
【摘要】 采用PCR技术对草地藏系绵羊心脏脂肪酸结合蛋白基因第2内含子进行了扩增,扩增产物经克隆和测序显示,其长度为1 893bp,该基因片段与GenBank中普通绵羊和猪的H-FABP基因第2内含子分别有99%和66.4%的同源性。实验还对22只草地藏系绵羊H-FABP基因第2内含子中174 bp的序列进行了PCR-SSCP分析,结果未检测到多态性。
【Abstract】 The second intron of heart fatty acid binding protein(H-FABP) gene of Tibetan sheep in Pasture was amplified by PCR method.The amplified PCR product was cloned and sequenced.The results showed that the length of fragment of H-FABP was 1 893 bp and this fragment had 99% and 66.4% of homology with the second intron of H-FABP gene of common sheep and porcine in GenBbank respectively.In addition,A 174bp-fragment of the second intron of H-FABP gene was assayed in 22 DNA samples from Tibetan sheep by PCR-SSCP,but no polymorphism was observed.
