鸟氨酸脱羧酶表达水平与溃疡性结肠炎病变时相的关系

【摘要】 【目的】观察鸟氨酸脱羧酶(ODC)表达水平与不同时相溃疡性结肠炎(UC)模型大鼠结肠病理变化的关系。【方法】Wistar大鼠60只,随机分为正常组(12只),模型1、2、3组(各16只),以90 mg/kg三硝基苯磺酸(TNBS)结肠注射复制UC模型,分别于实验第10、20、30天处死模型1、2、3组大鼠,正常组于第30天处死;分别对各组大鼠结肠进行大体和组织病理学评分,并采用免疫组化法检测结肠ODC的表达。【结果】随着实验周期的延长,模型大鼠结肠病变从急性炎症、组织坏死向慢性炎症、溃疡修复进展,ODC在正常结肠粘膜低表达,在溃疡形成、组织坏死阶段的表达略高于正常组,在溃疡修复、细胞大量增生时ODC表达明显增强,在病灶修复处于稳定阶段时ODC表达有所回落。【结论】不同时相UC模型大鼠结肠ODC表达变化特点与结肠病理变化特点相符。

【Abstract】 【Objective】 To investigate the relationship between ornithine decarboxylase(ODC) expression and pathological changes in ulcerative colitis(UC) at different periods.【Methods】Sixty Wistar rats were randomizedinto normal group (n=12) and model groups 1,2 and 3(n=16 in each group).UCanimal models in model groups 1,2 and 3 were induced bycolonicinjectionof 90 mg/kg TNBS(2,4,6-trinitrobenzensulfonic acid),and were executedrespectively in 10th,20thand 30thday after the modeling.The rats in the normal group were executed in 30thday after the modeling.Scoring of macroscopical features and pathological features was carried out in different groups and ODCexpressionin the colon was detected byimmunohistochemistry assay.【Results】With the prolongation of the experimental period,pathological changes in UC rats developed from acute inflammation and tissue necrosis to chronic in flammation and ulcer recovery.ODC was expressedinthe colon of normal rats,and the level was higher inthe period of ulceration and tissue necrosis in the model groupthanthat inthe normal group,increased obviouslyinthe period of ulcer recovery and cell proliferation,and slightly decreasedinthe stable stage of ulcer recovery.【Conclusion】ODCexpressionlevel inthe colon of UC rats varies inthe different pathological stages,andthe changes of ODCexpression are accorded withthe pathological characteristics.
