Bmp-2对牙周膜细胞增殖和碱性磷酸酶活性的影响
Impact of recombinant human bone morphogenetic protein-2 on proliferation and alkaline phosphatase activity of human periodontal ligament ceils in vitro
【摘要】 目的探讨骨形成蛋白-2(BMP-2)对人牙周膜(PDL)细胞增殖及碱性磷酸酶(ALP)活性的影响。方法通过体外细胞培养技术,用噻唑盐(MTT)法检测BMP-2刺激前后人PDL细胞增殖的情况,用酶动力学方法检测ALP活性。结果BMP-2可明显促进PDL细胞增殖,在实验的5d范围内,浓度在25~400μg/ml时呈浓度-时间依赖关系,最佳效应浓度为200μg/ml(比对照组增加了193.4%),最佳效应时间均为3d。BMP-2可显著增强PDL细胞的ALP活性,25μg/L作用下ALP活性已经比对照组差异有显著性(P<0.01),浓度为100μg/L时效果最佳(P<0.001),从第2d起,能增强人PDL细胞的ALP活性,第5d时作用趋于缓和。结论BMP-2能促进人PDL细胞的分化和增殖,能促进细胞ALP的活性。
【Abstract】 Objective To evaluate biological effects in vitro of recombinant human bone morphogenetie protein-2 on the prolif- eration and alkaline phosphatase(ALP)activity of human periodontal ligament(PDL)cells.Methods PDL cells were cultured for 1,2,3,4,5 days in DMEM supplemented with 1% FBS along(control),or containing BMP-2(25,50,100,200 and 400μg/ml). Proliferation of the PDL eells was measured by MTT colorimeteric assay.Alkaline phosphatase activity was determined by enzyme ki- netic method.Data was analyzed by ANOVA.Results BMP-2 at concentration of 25 to 400μg/ml showed a significant effects. BMP-2 promoted PDL cells growth in a dose-and time dependent manner.The most optimal coneentration of BMP-2 was 200μg/ ml(P<0.05).BMP-2 significantly increased ALP activity in PDL cells.Conclusion BMP-2 can promote the proliferation and significantly increased ALP activity of PDL cells.The data suggest that BMP-2 may be valuable in promoting the proliferation of periodontal connective tissue.
