扣囊复膜酵母菌细胞中海藻糖提取和精制工艺的研究

【摘要】 讨论几种海藻糖的提取方法,着重对比热蒸馏水法和热碳酸钠法提取海藻糖的优缺点。实验结果表明碳酸钠溶液提取得到的海藻糖量较多,但是提取液中的碳酸钠给后续的提纯工作带来很大的麻烦。用蒸馏水提取海藻糖成本低,无毒性,不需要溶剂回收设备,后续的提纯工作简单,得到的晶体纯度高。

【Abstract】 Several methods were discussed for trehalose extraction from cells of Saccharomycopsis fibuligera All,especially the hot distilled water extraction method and the hot sodium carbonate extraction method.We found that the hot distilled water extraction method was most suitable for trehalose extraction from the yeast cells and when the ratio of wet cell weight to the volume of distilled water was 1∶8,the trehalose yields were the highest.After ultrafiltration by 5 kDa cutoff membrane,desalinization and decoloration by ion chromatography and concentration by vacuum rotary evaporation,trehalose crystalline was obtained from the extract by using absolute ethanol.Analysis of TLC and HPEAC showed that the purified sample contained only trehalose which was identical to standard trehalose from Sigma.
