巨噬细胞高糖培养液促进肾小球系膜细胞增殖

【摘要】 目的分析高糖环境下巨噬细胞释放的因子在糖尿病肾病病理环节中的意义。方法Griess法检测高糖作用下的巨噬细胞上清中NO的含量;ELISA法检测条件培养液中TNF-α的含量;MTT法检测肾小球系膜细胞的增殖。结果12.5~50mol/L高浓度的葡萄糖体外作用24h、48h均能刺激大鼠腹腔巨噬细胞产生NO;TNF-α量显著增加(P<0.01)。大鼠巨噬细胞在含12.5mol/L葡萄糖的DMEM培养液中培养24h,其上清明显促进系膜细胞增殖的(P<0.01)。结论高糖环境中巨噬细胞释放NO,其上清液中NF-α量显著增加(P<0.01)等细胞因子,含有这些因子的细胞培养液明显促进系膜细胞增殖,提示高糖环境下的巨噬细胞是糖尿病肾病的发病的主要病理环节。

【Abstract】 OBJECTIVE To analyze the significance of the factors released by macrophage in high glucose environment in the pathology of diabetic nephropathy.METHOD Griess method was used to determine the NO content in macrophage clear supernatant.ELISA method was used to determine TNF-α content in culture fluid.MTT method was used to observe the hyperplasia of mesangial cell.RESULT In vitro action of 12.5 mol/L~50mol/L high concentration of glucose for 24 hours and 48 hours could stimulate the macrophage in rat abdomen to produce NO and TNF-α content increased markedly(P<0.01).The clear supernatant of rat macrophage cultivated in DMEM culture fluid containing 12.5 mol/L glucose could markedly promote the hyperplasia of mesangial cell(P<0.01).CONCLUSION The TNF-α in the clear supernatant of NO released by macrophage in high glucose environment increase significantly(P<0.01).The cell culture fluid containing these factors can significantly increase the hyperplasia of mesangial cell,suggesting that macrophage in high glucose environment is the main pathological factor in the pathogenesis of diabetic nephropathy.
