三种罗布麻干燥叶片基因组DNA的提取方法
Isolation of Genomic DNA From Dried Leaves of Three Kinds of Luobuma
【摘要】 采用Qiagen的植物基因组DNA提取试剂盒并加以改进,首次从不同季节采收、不同干燥方法保存的三种罗布麻(罗布红麻、大叶白麻和白麻)干燥叶片中提取出基因组DNA.提取的基因组TE溶解液的OD260/OD280值多在1.7-1.9范围内,电泳呈现单条带,核基因组的ITS片段、叶绿体基因组的trnL内含子和trnL-F非编码区片段的PCR扩增条带清晰并能进行DNA测序,这表明基因组DNA质量较好,可以满足后续分子生物学实验的要求.本研究为罗布麻等中药材的进一步分子生物学研究提供了必要的技术基础.
【Abstract】 Genomic DNA was first isolated from dried leaves of Luobuma by the modified method of the Qiagen DNeasy plant mini kit.Results showed that by this method genomic DNA could be isolated from Luobuma leaves of three species being collected at different seasons and being preserved by different methods.The OD260/OD280 ratios of the isolated DNA were in the range of 1.7~1.9,and only one band presented in agarose gels;clear PCR bands of ITS rDNA and trnL intron,trnL-F non-coding cpDNA could be sequenced.All the above revealed that the isolated DNA was complete and it could fulfill further demand for molecular biological experiment.
