斜纹夜蛾信息素结合蛋白cDNA片段的克隆及序列分析
Molecular cloning and sequence analysis of two cDNA fragments of pheromone binding protein in common cutworm,Spodoptera litura(Fabricius)
【摘要】 利用反转录多聚酶链式反应(RT-PCR)技术从斜纹夜蛾(Spodoptera litura(Fabricius))雄蛾触角扩增得到2个分别为275 bp和281 bp的信息素结合蛋白(PBP)cDNA片段:SlitPBP1和SlitPBP2,其分别由92个氨基酸残基和94个氨基酸残基组成,并且具有5个保守的半胱氨酸位点(全序列应有6个)。通过在GenBank中进行序列的同源性比较,表明这2个序列与已知几种夜蛾科昆虫的PBP氨基酸序列具较高的同源性。
【Abstract】 By reverse transcription-polymerase chain reaction(RT-PCR)method with a pair of degenerate primers,two cDNA fragments of pheromone binding protein(PBP)gene,SlitPBP1 and SlitPBP2,were cloned and sequenced from the antennae of male Spodoptera litura.SlitPBP1 and SlitPBP2 contained 275 bp and 281 bp,and with deduced amino acid sequences of 92 residues and 94 residues including five conservative Cys locus(six conservative Cys locus in full sequence),respectively.The sequence analysis indicated that the deduced amino acid sequences of the cDNA fragments shared high identity with PBP gene sequences from other previously reported moths.
