卡那霉素对不同小麦品种的效应及其在遗传转化筛选中的应用
Effect of Kanamycin on Different Cultivars and Its Application in the Selection of Transgenic Wheat
【摘要】 为建立利用卡那霉素筛选遗传转化小麦的有效体系,采用不同浓度的卡那霉素对小麦西农1376、郑麦9023、豫麦18、小偃22种子及幼苗进行筛选处理,并较为系统地研究了不同品种、不同时期及西农1376T1植株对卡那霉素的反应。结果表明,不同小麦品种幼苗期对卡那霉素的抗性存在显著基因型差异,同一品种存在不同的敏感时期和敏感浓度。西农1376幼苗的适宜筛选时期为三叶一心前,用20000 mg/L卡那霉素可对西农1376小麦转基因后代植株有效进行初步筛选。
【Abstract】 To establish the effective selecting system by using kanamycin in transgenic wheat,the response of 4 wheat varieties,Xinong 1376,Zhengmai 9023,Yumai 18,and Xiaoyan 22 to different concentrations at different culture phase was studied and the kanamycin’s effect on the T1 of Xinong 1376 was evaluated.The results indicated that the sensitivity of seedling to kanamycin was significantly different among these varieties.However,the resistance of kanamycin varied in different concentrations and at different seedling stage.The optimal screening stage of Xinong1376 was before three leaves.20 000 mg/L kanamycin was effective for preliminary screening of transgenic progeny plants of Xinong1376 wheat.
