麻风病患者皮损中分离出的分枝杆菌的分子鉴定
Identification of mycobacterial strains isolated from skin lesions of leprosy patients by molecular methods
【摘要】 目的:在分子水平上鉴定麻风病患者皮损中分离培养出的抗酸杆菌。方法:首先对麻风病患者皮损中分离出的9株分枝杆菌的hsp65基因进行PCR扩增;其次,对其PCR扩增产物进行酶切,然后,对该9株菌的PCR-限制性片段长度多态性(RFLP)分析的酶切带型进行分析;最后,再据上述初步鉴定结果,用特异性引物进行PCR扩增予以证实。结果:PCR-RFLP获得的酶切带型皆与细胞内分枝杆菌的酶切带型一致;用细胞内分枝杆菌的特异性引物分别对该9株临床分离株的模板DNA进行PCR扩增后,均获得271bp大小的特异性阳性条带。结论:麻风病患者皮损中分离出的9株分枝杆菌,其基因型表现属于细胞内分枝杆菌。
【Abstract】 Objective: To identify mycobacterial strains isolated from skin lesions of leprosy patients. Methods: Firstly, polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of the hsp 65 gene was performed on 9 mycobacterial strains isolated from skin lesions of leprosy patients. Secondly, PCR with primers specific to Mycobacterium intracellulare was applied to these mycobacterial strains. Results: The PCR-RFLP patterns of the 9 mycobacterial strains were all similar to those of M.intracellulare. The positive bands of 271 bp were got by PCR with primers specific to M.intracellulare. Conclusion: The 9 mycobacterial strains belong to M. intracellulare.
