金葡菌肠毒素A和B诱导BALB/c鼠皮肤炎症的实验研究

【摘要】 目的:探讨金葡菌肠毒素A(SEA)和金葡菌肠毒素B(SEB)在特应性皮炎发病机制中的作用。方法:使用100μg/mL浓度的SEA或SEB溶液分别斑贴致敏BALB/c鼠,以生理盐水为阴性对照,ELISA测定血清总IgE水平,观察和比较各组小鼠皮肤组织病理改变,半定量RT-PCR法测定致敏局部皮肤中白介素-4(IL-4)、白介素-5(IL-5),干扰素-γ(IFN-γ)mRNA表达水平。结果:SEA组及SEB组小鼠组织病理未见明显炎症性变化,各组小鼠间血清IgE水平无明显差异,IL-4、IL-5、IFN-γmRNA表达水平无统计学差异(P>0.05)。结论:100μg/L的SEA或SEB用斑贴致敏的方法不能在BALB/c鼠诱发具有特应性皮炎特征的皮损。

【Abstract】 Objective: To investigate the effects of staphylococcal enterotoxin A (SEA) and B (SEB) in the pathogenesis of atopic dermatitis. Methods: 100 μg/L SEA or SEB patch test was used to sensitize the skin of BALB/c mice and saline was used as negative control. The skin samples were obtained after sensitization and challenge. The histological change was observed and evaluated. Serum IgE level was measured by ELISA. Expression of IL-4, IL-5 and IFN-γ mRNA was detected by semi-quantified RT-PCR. Results: The experiment group did not show local skin inflammation. Serum IgE level and the levels of IL-4, IL-5 and IFN-γmRNA in the site of sensitized skin showed no statistically significant difference (P>0.05). Conclusion: Skin sensitization with 100 μg/mL SEA or SEB solution can not induce the inflammatory characteristics of atopic dermatitis in BALB/c mice.
