番茄组织再生及其遗传转化体系的优化
Regeneration and Optimization of Transformation System in Tomato
【摘要】 本研究首先以番茄10个自交系为实验材料,分别研究其不同激素组合对出愈和诱芽的影响,筛选出番茄自交系外植体再生的最佳激素组合为MS+ZT 2.0mg/L+IAA 0.5mg/L。试验结果表明,10个番茄自交系间外植体出愈率和诱芽率均存在较大的差异,说明基因型不同的自交系材料,其内源激素水平有所差异。采用正交实验设计,利用上述筛选出的再生体系对农杆菌侵染条件进行优化。结果表明,预培养2~3d、农杆菌稀释30倍、侵染时间20min,共培养2d,即可达到最佳的遗传转化效果,使番茄植株遗传转化效率达到最大化。
【Abstract】 10 tomato lines tested were used to study effects of different hormones combination on callus and Shoot regeneration.The optimistic combination selected is MS+ZT 2.0mg/L+IAA 0.5mg/L.The results indicated the rate of callus and Shoot regeneration of explants in 10 tomato lines are different.It demonstrates endogenesis hormones of different genotype lines are different.Using regeneration system selected above-mentioned,Agrobacterium transformation factors were tested.The most transformation effect is pre-culture 2~3 days,Agrobacterium diluted to 30 times,infected time 20 minutes and co-culture 2 days.
