座壳孢次生代谢物对Raji细胞增殖的影响

【摘要】 座壳孢(Aschersonia)是一类重要的虫生真菌成员,其次生代谢产物具有杀虫、抑菌等生物活性。本研究选取两株座壳孢菌(WYYS-6和WYSD-16),大量培养后从其培养液中萃取次生代谢产物,将座壳孢菌的次生代谢产物与Burkitt淋巴瘤细胞系Raji共孵育,然后应用四唑蓝[3-(4,5-双甲基-2-噻唑)-(2,5)-苯基溴化四氮唑蓝,MTT]检测方法研究了次生代谢产物在浓度为1mg/ml(稀释10倍)和0.1mg/ml(稀释100倍)下对淋巴系统恶性肿瘤Raji细胞增殖的影响。培养48h后,MTT检测显示1mg/ml的座壳孢菌WYYS-6次生代谢产物可以明显抑制Raji细胞增殖,而WYSD-16代谢产物(1mg/ml和0.1mg/ml)和0.1mg/ml的WYYS-6次生代谢产物对Raji细胞增殖未呈现出显著的抑制作用。

【Abstract】 Aschersonia is an important member of entomogenous fungi and its secondary metabolites show insecticidal and antibacterial activities.Two Aschersonia strains including WYYS-6 and WYSD-16 were chosen for mass production and their secondary metabolites were extracted with fractionation of organic extracts.Proliferation of human lymphoma cell line Raji incubated with different concentrations(1mg/ml and 0.1mg/ml) of Aschersonia secondary metabolites was evaluated with MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2h-tetrazoliumbromide)assay,respectively.After incubation with the secondary metabolites for 48 hours,MTT assay demonstrated that Aschersonia WYYS-6(1mg/ml) metabolites had extremely significant inhibitory effects on the proliferation of Raji cell.However,secondary metabolites of Aschersonia WYSD-16 and WYYS-6(0.1mg/ml) could not significantly inhibit Raji cell proliferation.
