携带STAT3基因的siRNA表达质粒的构建与鉴定
CONSTRUCTION AND IDENTIFICATION OF STAT3 GENE SIRNA EXPRESSION PLASMID
【摘要】 目的:构建含人STAT3基因不同位点的一系列siRNA真核表达质粒pSUPER,并进行测序,为以STAT3为靶点的肿瘤基因治疗研究奠定基础。方法:将人工合成的针对STAT3基因2个不同位点经基因重组定向克隆插入到真核表达质粒pSUPER中,通过PCR检测及质粒测序确定重组结果。结果:各对mRNA序列均按正确方向插入pSUPER质粒。结论:成功构建了含人STAT3基因2个不同位点的siRNA真核表达质粒。
【Abstract】 Objective:To construct a series of eukaryotic expression plasmid pSUPER which can transcript siRNA against human STAT3 gene for future study of the cancer gene therapy.Methods:Two pairs of different sites of STAT3 gene were synthesized.They were cloned into the eukaryotic expression vector pmU6 with definite direction.The recombinated plasmids were identified by PCR test and DNA sequence.Results:The siRNA sequences were successfully inserted into the eukaryotic expression vector pmU6.Conclusion:Two siRNA transcripting plasmids pSUPER are successfully constructed.
