人脐血单核细胞诱导分化为神经样细胞的实验研究

【摘要】 目的探讨人脐血单核细胞向神经细胞分化的潜能,为其治疗神经系统疾病提供实验依据。方法采用HES法分离脐血单核细胞(UCBMNCs),L-DMEM培养液原代和传代培养UCBMNCs,碱性成纤维细胞生长因子诱导细胞向神经细胞分化,应用流式细胞仪和免疫组化方法分析细胞中nestin、NSE阳性表达情况。结果原代UCBMNCs中有少量细胞表达nestin,阳性率为3.85%±0.88%;经诱导分化后原代(P0)和第10代(P10)细胞形态向神经样细胞转变:NSE阳性表达率分别为25.8%±4.6%、89.3%±8.5%;nestin阳性表达率分别为27.8%±5.8%和86.7%±6.8%。结论UCBMNCs具备神经样细胞分化潜能。

【Abstract】 Objective To research the differentiation potential of induccing human umbilical cord blood mononuclear cells(UCBMNCs) into neuron-like cells in vitro. Methods HES was used to separate mononuclear cells from human umbilical cord blood. L-DMEM medium was used to culture UCBMNCs,bFGF was used to induce UCBMNCs differentiation, flow cytometer and immunohistochemistry were used to analyze the differentiation results.Results A few cells in primary UCBMNCs expressed nestin, the rate was 3.85%±0.88%. After being induced,the P0 and P10 cells tended to differentiate into neuron-like cells,the rates of NSE positive cells in P0 and P10 were 25.8%±4.6% and 89.3%±8.5%, the nestin positive rates were 27.8%±5.8% and 86.7%±6.8% respectively.Conclusion UCBMNCs possess the potential to differentiate into neuron-like cells.
