氯化三乙基锡对大鼠C6脑胶质瘤细胞增殖和细胞周期的影响
Effect of triethyltin chloride on proliferation and cell cycle in rat C6 glioma cells
【摘要】 目的探讨氯化三乙基锡(triethyltin chloride,TETC)对体外培养大鼠C6脑胶质瘤细胞增殖和细胞周期的影响。方法采用MTT法检测0.5、1.0和2.0μmol/L TETC对体外培养大鼠C6脑胶质瘤细胞作用24h和48h后细胞增殖的影响、采用细胞形态学观察及流式细胞术(FCM)方法检测0.5、1.0和2.0μmol/L TETC对体外培养大鼠C6脑胶质瘤细胞作用48h后细胞增殖和细胞周期的影响。结果MTT法及细胞形态学观察均检测到0.5、1.0和2.0μmol/L TETC在体外均可抑制C6细胞增殖,C6细胞增殖率存在着剂量依赖性和时间依赖性降低趋势;TETC可将C6细胞阻滞在G0/G1期。结论TETC可抑制体外培养大鼠C6胶质瘤细胞增殖,其机制可能与C6细胞周期阻滞在G0/G1期有关。
【Abstract】 Purpose To study the effects of triethyltin chloride (TETC) on proliferation and cell cycle of cultured rat C6 glioma cells. Methods MTT assay was performed to evaluate the effect of 0.5, 1.0 and 2.0 μmol/L TETC on proliferation of rat C6 glioma cells for 24 h and 48 h. Optical microscope and flow cytometry were employed for determining the proliferatiing effect and the cell cycle distribution of C6 glioma cells treated by 0.5, 1.0 and 2.0 μmol/L TETC for 48 h, respectively. Results 0.5, 1.0 and 2.0 μmol/L TETC could inhibit the proliferation of cultured C6 glioma cells in vitro performed by both MTT assay and cell morphological observation, and the proliferative rate decreased in a dose-dependent and time-dependent manner. In addition, the cell cycle of C6 glioma cells was arrested at G0/G1 phase. Conclusions TETC inhibits the proliferation of cultured rat C6 glioma cells in vitro, the mechanism of which may be involved in cell cycle arrest of the cells.
