高血压患者血小板聚集功能与纤维蛋白原基因多态性的相关性研究
The relationship between platelet aggregation function and Bβ-fibrinogen gene-455G/A polymorphism in hypertensive patients
【摘要】 目的观察高血压患者血小板聚集功能变化是否与Bβ纤维蛋白原(Fib)基因启动子区-455G/A多态性有关。方法选取108例未经治疗的高血压患者,采用全血电阻抗法测定二磷酸腺苷(ADP)及花生四烯酸(AA)诱导的血小板聚集功能,并用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)方法测定BβFib基因启动子区-455G/A多态性,将其分为GG组和GA+AA组,比较两组血小板聚集功能变化。结果GA+AA组Fib浓度高于GG组(P<0.05),ADP诱导的血小板聚集能力GA+AA组低于GG组(P<0.05);而AA诱导的血小板聚集能力呈现GA+AA组高于GG组的趋势(P>0.05)。结论ADP诱导的血小板聚集功能与BβFib基因启动子区-455G/A多态性有关,GG基因型患者血小板聚集功能强于AA及GA基因型患者。但AA诱导的血小板聚集功能与其的关系呈现相反的趋势。
【Abstract】 Objective To investigate the relationship between platelet aggregation function and Bβ-fibrinogen gene-455G/A polymorphism in hypertensive patients.Methods Platelet aggregation function induced by ADP and arachidonic acid(AA) was determined by whole blood impedance method in 108 hypertensive patients.The-455G/A polymorphism of Bβ-fibrinogen was screened by PCR-RFLP.Platelet aggregation function was analyzed for groups genotype GG and genotype(GA+AA).Results Fibrinogen concentration in group GA + AA was higher than that in group GG(P<0.05).Platelet impedance(ohm) induced by ADP in group GG was higher than that in group GA + AA(P<0.05).Platelet impedance induced by AA in group GA+AA was higher than that in group GG(P>0.05).but there was no statistical difference.Conclusion Platelet impedance(ohm) induced by ADP correlates with the-455G/A polymorphism on the promoter region of Bβ-fibrinogen gene.Platelet aggregation function in hypertensive patients with genotype GG is stronger than that in patients with A-455 allele,while there is a contrary tendency for platelet aggregation function induced by AA.
