三七总皂苷微乳对小鼠模型的抗皮肤衰老作用
Anti-aging effect of microemulsion containing Panax notoginseng on skin
【摘要】 目的观察三七总皂苷微乳经皮给药抗皮肤衰老作用。方法昆明种小鼠每日颈背部皮下注射D-半乳糖(1 000 mg.kg-1.d-1)造成小鼠衰老模型,同时背部皮肤涂三七总皂苷微乳42 d后,测定小鼠背部皮肤组织匀浆中超氧化物歧化酶(SOD)、过氧化脂质代谢产物丙二醛(MDA)及皮肤羟脯氨酸含量;同时,对皮肤切片进行组织学病理形态观察及计算机图像定量分析。结果与对照组相比,三七总皂苷微乳使D-半乳糖所致的衰老模型对照组小鼠真皮厚度(594.11±156.96)μm较制剂组(848.29±123.57)μm显著变薄(P<0.01),胶原纤维增加,排列紧密,弹力纤维总面积也显著增加(P<0.01);皮肤中SOD活力(21.05±0.65)U.mg-1较模型组(14.51±1.8)U.mg-1及对照组(18.60±1.92)U.mg-1均显著提高;MDA含量(2.29±0.51)nmol.mg-1显著较模型组(3.88±1.62)nmol.mg-1和对照组(3.03±0.95)nmol.mg-1减少;羟脯氨酸含量(18.9±2.01)mg.g-1显著高于模型组(15.47±1.62)mg.g-1和对照组(16.4±1.73)mg.g-1。结论三七总皂苷微乳经皮给药对D-半乳糖所致的衰老模型小鼠皮肤有显著的抗衰作用。
【Abstract】 AIM To investigate the anti-aging effect of Panax notoginseng microemulsion on skin.METHODS The aging mice model was established by subcutaneous administration with a dose of D-galactose(1 000 mg·kg-1·d-1)to female mice for 6 wk,at the same time,the skin of back of mice were shaved topically and applied with microemulsion.After 42 d,the SOD activity,the malondialdehyde(MDA) and hydroxyproline contents in skin were determined.Meanwhile,histochemical standards relating to aging were measured and morphologic alterations of surplus dorsal skin were observed under microsecope and analyzed.RESULTS In contrast with the preparation group(848.29±123.57)μm,control group showed that the thickness of dermis(594.11±156.96)μm was significantly reduced(P<0.01),and the content of elastic fibers was increased(P<0.01) and accumulated and tangled.The activity of SOD(21.05±0.65) U·mg-1 and content of hydroproline(18.9±2.01)mg·g-1 were significantly decreased compared with control group [SOD(14.51±1.8)U·mg-1,hydroproline(15.47±1.62)mg·g-1]and blank group [SOD(18.60±1.92)U·mg-1,hydroproline(16.4±1.73)mg·g-1],whereas the content of MDA was significantly increased.CONCLUSION Microemulsion containing Panax notoginseng has obvious anti-aging effects on aging mice’s skin.
