低频脉冲磁场对脑缺血再灌注损伤大鼠神经元保护作用的研究

【摘要】 目的研究低频脉冲磁场对脑缺血再灌注损伤大鼠海马神经元的保护作用。方法将24只SD大鼠随机分成磁辐射组和对照组(每组12只),应用改良的Pulsineli法制作大鼠全脑缺血再灌注模型;将磁辐射组大鼠头部置于低频脉冲磁场(20mT、10Hz)辐射45min,每天1次,连续4d。进行脑电图监测,用Nissl染色及免疫组化方法检测海马神经元数及caspase-3蛋白阳性细胞数。结果磁辐射组脑电图振幅[(10.27±1.12)μV]和频率[(10.06±1.02)Hz]较对照组[(8.95±1.04)μV、(8.62±0.94)Hz]显著增加(均P<0.05);磁辐射组海马神经元数[(29.02±1.32)个/高倍镜视野]较对照组[(21.25±1.06)个/高倍镜视野]增多,磁辐射组caspase-3蛋白阳性细胞数[(21.33±1.32)个/高倍镜视野]较对照组[(28.34±1.10)个/高倍镜视野]减少,两组相比差异具有统计学意义(均P<0.05)。结论低频脉冲磁场可以减轻脑缺血再灌注后海马神经元的损伤及凋亡,对脑缺血再灌注损伤有保护作用。

【Abstract】 Objective To investigate the protective effects of low-frequency pulsed electromagnetic field (LFPMF) on neuron following cerebral ischemia-reperfusion injury in rats.Methods 24 adult male Sprague-Dawley rats were randomly divided into irradiation group (n=12) and control group (n=12). The global cerebral ischemia-reperfusion models of rats were produced by modified Pulsineli method. Rats in irradiation group were exposed to 20 mT intensity LFPMF of 10 Hz for 45 minutes per day for 4 days. The electroencephalogram (EEG) was recorded by powerlab system. Nissl staining was used to perform quantitative analysis of neuron number and immunohistochemistry staining provided the information of the distribution of positive cells to caspase-3.Results Compared with the control group, the amplitude of EEG was significantly higher in irradiation group [(10.27±1.12)μV versus (8.95±1.04)μV] (P<0.05), and the frequency was also faster [(10.06±1.02)Hz versus (8.62±0.94)Hz] (P<0.05). The number of neuron of hippocampus in irradiation group [(29.02±1.32)cells/HP] was more than that of control group [(21.25±1.06)cells/HP] (P<0.05), however, the number of positive cells staining to caspase-3 was fewer than control group [(21.33±1.32)cells/HP versus (28.34±1.10)cells/HP](P<0.05).Conclusions LFPMF may restrain the damage and apoptosis of the neuron in global cerebral ischemia-reperfusion injury, which is involved in the mechanisms of protective effect of LFPMF.
