肺癌患者GST-π基因表达的临床意义

【摘要】 目的探讨肺癌组织谷胱甘肽-S-转移酶-π(GST-π)基因表达及其对化疗疗效的影响。方法采用逆转录多聚酶链反应对60例经纤维支气管镜活检肺癌组织和20例非癌良性肺组织进行GST-πmRNA定量检测,肺癌患者行至少2个周期的铂制剂化疗。结果肺癌组织GST-πmRNA表达阳性率(45.0%)明显高于良性肺组织(15.0%,P<0.05),腺癌和鳞癌阳性率较高,分别为64.7%和45.5%,GST-πmRNA表达与肿瘤组织病理类型有相关性(P<0.05),GST-πmRNA阴性表达组化疗有效率(63.6%)显著高于阳性组(18.5%,P<0.05),组织GST-πmRNA表达与化疗疗效有相关性(P<0.05)。结论肺癌组织恶变过程中GST-πmRNA基因表达水平增加,GST-π可以作为肺癌耐药的预后因子,GST-π表达对临床化疗疗效判断有一定指导意义。

【Abstract】 Objective To investigate the GST-πgene expression and the instructive action to the chemotherapy in patients with lung cancer.Methods The GST-πof 60 lung cancer cases and 20 noncancer cases were detected by reverse transcription- polymerase chain reaction (RT-PCR).60 cases with lung cancer were treated with platin-based chemotherapy for more than two periods.Results GST-πgene expression in lung cancer tissues was higher than that of the noncancer tissues(P<0.05).The positive expression rate of GST-π in adenocarcinoma and squamous carcinoma was 64.7% and 45.5%,respectively.GST-πgene expression was correlated to pathological type(P<0.05).The response rate to the chemotherapy of the GST-πnegative group (63.6%) was significantly higher than that of the positive group(18.5%,P<0.05).Correlations were noted between the GST-πgene expression and the therapy effect(P<0.05).Conclusion The genetic expression of GST-π was increased in the cancerization course of lung cancer.GST-πcan be a tumormarker in diagnosing lung cancer.The GST-πgene expression has a significant instructive value in estimating the curative effect.
