【摘要】 目的研究白细胞介素3受体α mRNA(IL-3RαmRNA)在急性白血病(AL)中的表达及其在急性白血病中的临床意义。方法对80例急性白血病患者(包括56例初治急性白血病患者、9例复发患者和15例缓解患者),另外,15例健康人为正常对照组,KG-1α细胞株为阳性对照,应用半定量逆转录-聚合酶链反应检测各组IL-3RαmRNA水平。结果IL-3RαmRNA在初治AL患者中表达水平明显高与正常对照组(P<0.05);IL-3Rα mRNA在缓解组表达水平高于正常对照组,但差别无统计学意义(P>0.05),但缓解组表达水平低于初治AL组,且差别有统计学意义(P<0.05);复发组平均表达水平高于正常对照组(P<0.05);AL患者中42例可进行疗效评估,初治IL-3Rα mRNA阳性AL患者中分为高表达与低表达组,IL-3Rα低表达患者的缓解率明显高于高表达患者,两者比较差别有统计学意义(P<0.05);7例复发的AL患者,仅有1例用药后达到完全缓解(CR),其IL-3Rα mRNA表达均阳性;余6例均未达到CR。结论在急性白血病患者IL-3Rα mRNA有异常过度高表达;IL-3Rα mRNA高表达的患者缓解率低,表明与白血病的发病及疗效预后有关;可用来作为判断疗效预后的一新指标。更多还原

【Abstract】 Objective To investigate the expression of interleukin3 receptor alpha(IL-3Rα) chain in leukemia and the prognostic significance in acute leukemia(AL).Methods The expression of IL-3Rα mRNA was measured in 80 acute leukemia patients,including 56 de novo acute leukemia patients,9 replased patients,15 complete remission(CR) patients,besides,15 adults were selected as normal controls(NC) and KG-1α cell line was employed as positive control.All the subjects were measured by semi-quantity reverse transcription polymerase chain reaction(RT-PCR).Results The expression of IL-3Rα mRNA in de novo AL patients was significantly higher than that of NC(P<0.05);In CR patients IL-3Rα mRNA was higher than that in NC,but no significant difference(P>0.05),IL-3Rα mRNA in AL group was higher than that in CR group(P<0.05).The expression of IL-3Rα mRNA in AL was higher than that in NC(P<0.05).In relapsed group,IL-3Rα mRNA was higher than that in NC(P<0.05),an effect evaluation was made on 42 cases of patients.IL-3Rα+ patients were divided into IL-3Rαlo group and IL-3Rαhi group.In IL-3Rαlo group,the CR rate was significantly higher than that in IL-3Rαhi group(P<0.05).In relapsed group,only one of them achieved remission by combined chemotherapy,who was positive with IL-3Rα.The rest of them did not achieve remission.Conclusion IL-3Rα mRNA expresses widely in specimens of AL,IL-3Rα mRNA expression is associated with remission rate in AL.The patients with high level of IL-3Rα mRNA have a low remission rate,which is concerned with the leukemia morbidity and the curative effect prognosis,and may be taken as a new target for the judgment of the curative effect and prognosis.更多还原
