T淋巴细胞协同刺激分子表达异常与类风湿性关节炎患者免疫功能紊乱
Relationship between abnormality of costimulative molecular expression and imbalanced immune function in patients with rheumatoid arthritis
【摘要】 目的探讨类风湿性关节炎(RA)患者T细胞表面协同刺激分子表达与其发病的相关性及临床意义。方法采用流式细胞术分析RA患者外周血T淋巴细胞表面共刺激分子CD28、CD152和CD154的表达。结果RA患者外周血CD4+T细胞较正常对照组增加,分别为(41.86±7.67)%vs(32.20±6.51)%,CD8+T细胞减少,分别为(23.82±6.88)%vs(28.16±8.43)%,差异均有统计学意义(均P<0.05);协同刺激分子CD28在RA患者较对照组CD4+和CD8+T细胞亚群上分子的表达均减少分别为(9.81±1.36)%vs(42.13±0.59)%和(7.27±3.24)%vs(18.56±2.17)%,而CD152分子的表达均增加,分别为(0.46±1.21)%vs(0.13±0.42)%和(18.78±0.14)%vs(0.42±0.47)%,与对照组比较,差异均有统计学意义(均P<0.05);CD154分子在RA患者CD4+T细胞和CD8+T细胞上表达较正常对照组也均增加,分别为(3.64±2.61%)vs(2.37±1.05)%和(4.23±1.24)%vs(2.13±0.67)%,差异有统计学意义(P<0.05)。结论RA患者机体的免疫功能紊乱与T淋巴细胞表面协同刺激信号分子表达异常具有相关性。
【Abstract】 Objective To explore the relationship between expression of costimulative molecules on T cells in the patients with rheumatoid arthritis(RA) and their clinical significance. Methods Expressions of costimulative CD28,CD152 and CD154 on T lymphocytes were analyzed by flow cytometry. Results Imbalanced T subsets,increasing CD4+ T cells( 41.86± 7.67)% vs ( 32.20± 6.51)% and decreasing CD8+ T cells( 23.82± 6.88)% vs ( 28.16± 8.43)% were found in the peripheral blood of RA group compared to those of control group(all P< 0.05). Compared to those of control group,the expressions of CD28 on both CD4+T cell and CD8+ T cell in RA group decreased ( 9.91± 1.36)% vs ( 42.13± 0.596)%;( 7.27± 3.24)% vs ( 18.56± 2.17)%, but the expression of CD152 increased ( 0.46± 1.21)% vs ( 0.13± 0.42)%;( 18.78± 0.14)% vs ( 0.42± 0.47)%. The expression of CD28 decreased but the expression of CD152 increased,with significant difference (all P< 0.05).The expressions of CD154 on both CD4+ and CD8+ T cells in RA group increased compared to those of control group ( 3.64± 2.61%) vs ( 2.37± 1.05)%;( 4.23± 1.24)% vs ( 2.13± 0.67)%(P< 0.05). Conclusion Abnormal expressions of costimulative signal molecules on T lymphocytes lead to the imbalanced immune function in the patients with rheumatoid arthritis.
