口腔黏膜下纤维性变发病相关基因的功能分析
Analysis on the Oral Submucous Fibrosis-related Gene by cDNA Chip
【摘要】 目的:应用cDNA芯片技术研究口腔黏膜下纤维性变(oral submucous fibrosis,OSF)颊黏膜上皮组织和正常口腔颊黏膜组织基因的差异表达,并对差异表达基因进行gene ontology(GO)分析,为探讨OSF发病相关基因提供初步筛选资料。方法:对切取OSF和正常对照颊黏膜组织进行总RNA抽提,逆转录制备探针,纯化后与含有21571条基因的人类全基因组寡核苷酸芯片杂交,杂交后信号经扫描仪检测和计算机分析筛选OSF发病相关差异表达基因,并进行GO分析。结果:GO分析发现,2372条差异表达基因涉及细胞组成的基因群主要集中于细胞外基质和细胞骨架,与溶酶体囊胞、细胞小凹等蛋白相关基因多呈上调表达。结论:细胞外基质、细胞骨架、溶酶体囊胞、细胞小凹等蛋白相关基因群差异表达可能与OSF发病有关。
【Abstract】 Objective: To analyse oral submucous fibrosis-related gene by cDNA chips so as to investigate the mechanism of oral submucous fibrosis. Methods: We studied novel gene expression by cDNA chips, and then analysed the gene differently expression profile by Gene Ontology. The total RNA from the OSF tissues or control tissues were reversely transcribed to cDNA with the incoporation of fluorescent-labeled to prepare the hybridization probes. After hybridization, cDNA chips were scanned for the fluorescet intensity. OSF-related gene expression was screened through the gene ontology analysis of difference in gene expression profiles. Results: 2372differently expression cellular-compoent related genes mostly focused on the cytoskeleton, extracellular matrix, lytic valuole and caveola. Conclusion: The result showed that the differently expression gene, associated with the cytoskeleton, extracellular matrix, lytic valuole and caveola, may be the mechanism of oral submucous fibrosis.
