创伤性深静脉血栓形成中TNF-α、IL-1β、IL-6和IL-10的变化和作用
The Changes and Roles of TNF-α, IL-1β, IL-6 and IL-10 in Traumatic Deep Venous Thrombosis
【摘要】 目的探讨创伤后炎症因子在深静脉血栓形成中的变化和作用.方法采用血管钳夹+固定制动的方式建立创伤性深静脉血栓形成动物模型,根据观察时间和/或血栓存在状态将实验动物分为7组,分别获取血清标本,进行TNF-α、IL-1β、IL-6和IL-10ELISA测定;分别切取股静脉血管测定血栓质量并进行组织学观察.结果创伤性深静脉血栓形成中伴随急性炎症向慢性炎症转变的过程;血清中TNF-α、IL-1β、IL-6和IL-10在不同时相点、不同的血栓存在状态表达水平不同;血栓质量与TNF-α、IL-1β、IL-6和IL-10之间无相关性(P>0.05),但血栓质量与IL-1β/IL-10的比值存在正相关(Pearson系数=0.87,P=0.01<0.05).结论创伤后炎症反应与创伤性深静脉血栓形成之间存在密切的关系.监测IL-1β/IL-10的比值可以推测创伤后深静脉血栓的形成和预后.
【Abstract】 Objective To explore the changes and roles of inflammatory factors in deep venous thrombosis after trauma. Methods Rat traumatic deep venous thrombosis models were set up by clamping the femoral vein and fixing the bilateral hind limbs. According to observation times and/or the thrombosis these rats were divided into 7 groups. Serum samples were obtained and femoral vein thrombus mass were weighed from each group rat. Then, these acquired serum were observed by light microscope and TNF-α,IL-1β,IL-6 and IL-10 were assayed by ELISA. Venous blood vessel incised to determine the thrombus quality and conduct histological observation. Results There was a transformation process from acute inflammation to chronic inflammation in vein wall during the course of deep venous thrombosis. TNF-α,IL-1β,IL-6 and IL-10 in serum were not invariant at different observation times and different thrombosis conditions. There was no correlation between TNF-α,IL-1β,IL-6 or IL-10 and thrombus mass(all P>0.05)but there was a correlation of IL-1β/IL-10 ratio with thrombus mass (Pearson r=0.87, P=0.01<0.05). Conclusions There was a close dependability between inflammation and deep vein thrombosis after trauma. Monitoring the ratio of IL-1β/IL-10 can predict deep venous thrombosis and its prognosis after trauma.
