应用大孔吸附树脂分离纯化葶苈子总黄酮的研究

【摘要】 目的考察6种大孔树脂对葶苈子总黄酮成分的分离纯化性能,确定最佳工艺条件。方法以总黄酮吸附量、洗脱率、洗脱物总黄酮含量为指标,采用紫外-可见分光光度法进行含量测定。结果NKA-Ⅱ型树脂对葶苈子总黄酮具有较好的纯化分离能力,其最佳工艺条件为:葶苈子提取物浓度4g/mL(生药量),最大吸附量12.92mg/g,装柱径高比1∶12,吸附流速3mL/min,洗脱剂80%乙醇,洗脱量5倍柱体积,洗脱流速3mL/min。结论NKA-Ⅱ型树脂纯化分离葶苈子总黄酮的最佳工艺稳定高效,可推广用于生产。

【Abstract】 Objective To establish the purification and separation technology for total flavones from Semen Descuraniae by investigating 6 types of macroporous adsorption resin.Method The adsorption capacity,elution ratio and purify of total flavones were used as evaluating criteria.The UV-VIS spectrophotometric method was used in the determination of total flavones content.Results NKA-Ⅱresin had better property of purification and separation.The optimal technological conditions were: the concentration of the sample of Semen Descuraniae,4 g/ml(amount of crude drug); the maximum adsorption capacity for total flavones,12.92 mg/g;the optimal column ratio of diameter to height,1/12; the flow velocity of adsorption,3ml/min ; the eluting reagent, 80% ethanol(5 times of the volume of the resin);eluting velocity , 3ml/min.Conclusion The technology of separating and purifying total flavones from Semen Descuraniae with NKA-Ⅱwas stable and effective, and can be applied to manufacture.
