新型化学氧化法测定胆红素试剂的研制
Development of the reagents with new chemical oxidation method for measuring serum bilirubin
【摘要】 目的探讨建立一种新型化学氧化法测定血清总胆红素和直接胆红素的新方法。方法采用化学氧化法测定血清胆红素反应前后吸光度的变化,与标准品相比较,求得标本中总胆红素和直接胆红素的浓度。结果TBIL精密度试验批内高低浓度CV值为0.65%~1.56%;DBIL为0.82%~1.93%。批间TBIL为3.5%、1.71%及1.24%;DBIL批间为4.38%、2.77%及1.60%。线性范围:TBIL可达686μmol/L;DBIL可达400μmol/L;总胆回收率101.8%~107.6%,平均104.5%.DBIL回收率为99.1%~100.5%,平均为100.2%。与其他相关试剂比较TBILγ=0.9973~0.9992;DBILγ=0.9962~0.9994。溶血、脂浊和维生素C干扰试验表明:标本中Hb小于2.7g/L,TG小于5.94mmol/L。VitC小于0.1g/L,对TBIL和DBIL的测定基本无干扰。结论本试剂方法简便,结果准确、线性范围宽,试剂稳定,价格低廉。完全可用于检测临床标本。
【Abstract】 Objective To establish a new chemical oxidation method for measuring serum bilirubin. Methods Chemical oxidation method was applied for detecting serum total bilirubin(TBIL) and cholebilirubin(DBIL) concentrations according to the absorbance changes,and the results were analysed comparing with the standard controls. Results The intra-assay CVs of TBIL were 0.65% and 1.56% (n=15), with the mean values of 29.35 and 221.7μmol/L respectively, while that of DBIL were 0.82% and 1.93% (n=15), with the mean values of 9.70 and 119.76μmol/L respectively. The inter-assay CVs of TBIL were 3.50%, 1.71% and 1.24% (n=20), with the mean values of 15.67, 214.47 and 564.98μmol/L respectively, while that of DBIL were 4.38%, 2.77% and 1.60% (n=20), with the mean values of 5.18, 76.12 and 251.83μmol/L respectively. The linear ranges were up to 686μmol/L for TBIL and 400μmol/L for DBIL respestively. The average recoveries were 104.5% for TBIL and 100.2% for DBIL. The nitrite method was well correlated with Vanadate method and Diazo method. When hemoglobin level in serum was under 2.7g/L, Triglycerides under 5.94 mmol/L, or Vitamin C under 0.1g/L, the results showed no significant interference for measuring bilirubin. Conclusions This new chemical oxidization method is simple and stable, and it can be used in routine detections of serum total TBIL and DBIL.
