种子工艺优化对链霉菌702发酵产抑真菌生物活性物质的影响

【摘要】 采用单因素实验、正交设计对链霉菌702种子培养基及其培养条件进行了优化研究。优化后种子培养基配方为:蔗糖2.0%、玉米粉2.0%、黄豆饼粉1.0%、酵母膏0.4%、碳酸钙0.6%、豆油0.4%;优化的种子培养条件为:20 mL种子培养基/250 mL三角瓶、接种孢子悬液(T660 nm=80%)体积比为2.5%、起始pH为8.0、培养周期为48 h。链霉菌702菌种在优化的种子工艺上培养、发酵后的抑真菌物质生物效价达到983.62μg/mL,比优化前的770.23μg/mL提高了27.70%。

【Abstract】 The seed medium and the culture conditions of streptomyces 702 was studied by one-way experiments and orthogonal test.The optimum seed medium was sucrose 2.0%,cornmeal 2.0%,soybean power 1.0%,yeast extract 0.4%,CaCO3 0.6%,soybean oil 0.4%;the optimum seed culture conditions were as follows:20ml liquid seed culture medium/250 mL flask,the spore(T660 nm=80%)inoculation rate was 2.5%,the initial pH 8.0,culture time was 48 hours.Under the optimum seed fermentation technology,the yield of anti-fungus bioactive substance produced by Streptomyces 702 increased 27.7% than that of the original 770.23 μg/mL,and reached to 983.62 μg/mL.
