免疫介导的脊髓运动神经元损伤过程中细胞周期素依赖蛋白激酶5的表达及意义
CDK5 expression of spinal motor neuron in immune-mediated injury
【摘要】 目的:研究免疫介导的神经元损伤过程中细胞周期素依赖蛋白激酶5(CDK5)表达的特征,探讨其损伤机制。方法:建立免疫介导的脊髓前角运动神经元损伤的模型,利用免疫组化方法观察脊髓前角运动神经元CDK5及p35/ p25的表达特征。结果:对照组豚鼠脊髓运动神经元CDK5及p35/p25主要在细胞核及细胞膜下表达,胞质着色淡;注射牛脊髓前角匀浆而无肢体瘫痪的豚鼠,脊髓运动神经元CDK5及p35/p25表达在数量上增加,强度上增强;有明显肢体瘫痪的豚鼠CDK5及p35/p25免疫反应阳性神经元数量明显减少,部分胞质内无着色。结论:CDK5及p35/ p25在神经元胞质内的易位表达,在免疫介导的神经元损伤过程中具有重要的作用。
【Abstract】 Objective.To observe CDK5 and activator p35/p25 expressions and investigate the pathogenesis of motor neuron damage in immune-mediated injury.Methods:Experimental autoimmune gray matter disease (EAGMD) animal model was established and the expression features of CDK5 and activator p35/p25 in spinal motor neuron were observed by immunohistochemical methods.Results:Our data showed that CDK5 and activator p35/p25 were located under the membrane of spinal motor neurons in the control guinea pigs.The expressions of CDK5 and p35/p25 increased in the motor neuronal somata of spinal cord ventral horn and some proximal axons of the gray matter-immuned guinea pigs without limb paralysis.In contrast,CDK5 inmmunoreactive neurons in the spinal cord of gray matter-immunized guinea pigs with limb paralysis were rare.There was signifieant difference between the EAGMD and the control guinea pigs.Conclusion:The mislocation of CDK5 and activator p35/p25 may play an important role in immune-mediated mo- tor neuron injury.
