【摘要】 目的探索PI3K/PKB信号转导通路在胃癌发生发展过程中的作用及其与PTEN的相关性。方法对112例胃癌及76例正常胃粘膜组织采用免疫组织化学Envision二步法检测PI3K、PKB和PTEN蛋白、采用原位分子杂交法检测PTENmRNA。结果胃癌组PI3K及PKB表达均明显高于正常胃粘膜组(分别为χ2=9.994,P<0.05;χ2=15.823,P<0.001);随着癌浸润深度的加深(χ2=7.896,P<0.05;χ2=7.939,P<0.05)和淋巴结转移的出现(χ2=9.455,P<0.05;χ2=8.115,P<0.05)PI3K及PKB表达明显增强。而PTEN在胃癌中的表达明显低于正常胃粘膜(χ2=78.494,P<0.001);在胃癌浸润超过肌层组明显低于未超过肌层组(χ2=11.308,P<0.01);伴淋巴结转移组表达明显低于无淋巴结转移组(χ2=17.870,P<0.001)。胃癌组PTENmRNA表达明显低于正常胃粘膜组(χ2=53.959,P<0.001)。且PTEN与PI3K及PKB蛋白表达强度呈明显负相关(r=-0.354,P<0.001;r=-0.222,P<0.05)。结论PI3K/PKB信号转导通路和PTEN在胃癌发生发展过程中分别起促进和抑制作用,两者呈负相关,PI3K/PKB通路有望成为PTEN失活时治疗胃癌的靶点。更多还原

【Abstract】 Objective To explore relationship between the expression of PI3K/PKB signal pathway and PTEN on gastric cancer.Methods Detecting PI3K,PKB,PTEN proteins and PTEN mRNA of normal gastric mucosa and gastric carcinoma by immunohistochemistry EnVision method and molecular hybridization in situ method respectively.Results The expression of PI3K and PKB of gastric carcinoma increased than that of normal gastric mucosa(P<0.05).The expression of PI3K and PKB of non-running muscular layer group is obviously higher than that of running muscular layer group(P<0.05),and the results was semblable in lymphatic metastasis group contrasting with non-lymphatic metastasis group(P<0.05).Meanwhile,the expression of PTEN protein was significant lower in gastric carcinoma group,running muscular layer group and lymph-metastasis comparing with normal gastric mucosa group,non-running muscular laye group and non-lymph-metastasis group(P<0.05),and there is negative correlationn between PTEN and PI3K/PKB pathnay.The expression of PTEN mRNA of gastric carcinoma group is less than that of normal gastric mucosa group(P<0.001).Conclusion There is closely relationship between PTEN and PI3K/PKB signal pathway,PTEN and PI3K,PKB play an important role in carcinogenesis and development of gastric carcinoma and may become the treating target of gastric cancer.更多还原
