顺铂腹腔注射对小鼠卵巢相关基因表达谱的影响

【摘要】 目的:应用基因芯片技术研究顺铂(Clsplatin)腹腔注射给药对小鼠卵巢相关基因表达的影响,探讨顺铂对卵巢功能损伤的作用机制。方法:建立化疗损伤后卵巢功能低下小鼠模型,按一步法抽提顺铂给药后的小鼠及对照组小鼠卵巢组织的总RNA;经逆转录分别用Cy3、Cy5荧光标记,获得两组小鼠卵巢cDNA的探针,再与cDNA基因表达谱芯片杂交,结果由激光扫描仪扫描,并用计算机软件进行图像分析、标准化处理、ratio值分析和聚类分析。结果:顺铂给药后的小鼠与对照组小鼠卵巢组织基因表达谱分析发现,109个差异表达基因有19个基因低表达,90个基因高表达。结论:本研究从顺铂给药后的小鼠卵巢组织中筛选出大量的差异表达基因,说明这些基因可能参与顺铂对卵巢功能损伤的发生、发展过程,从而为化疗药物损伤性卵巢功能减退的诊治提供依据。

【Abstract】 Objective:To investigate the effect of ovarian related genes expression in mice intraperitoneally injected with cisplatin by microarray technology and to analysis the mechanism of ovary function induced by cisplatin.Methods:The model of ovarian dysfunction mice induced by chemotherapy was established.One-step method for extraction of total RNA of mice ovary tissue after administration of cisplatin and the ovarian tissue of the normal control group was used.After reverse transcription and fluorescent labeling by Cy3,Cy5,we get two groups of mouse ovarian cDNA probes and then hybridized with the cDNA gene expression profiles microarray hybridization.The result was scanned by the laser scanner and processed by software for image analysis,standardization,the ratio of value analysis,cluster analysis.Results:By analyzing the ovarian tissue gene expression after the administration of cisplatin,we found 109 differentially expressed genes,among which,there are 19 up-regulated expression genes and 90 down-regulated expression genes.Conclusion:The analysis of ovarian gene expression profiles of mice intraperitoneally injected by cisplatin based on cDNA microarray can screen considerable differential expression genes which may participate in the occurrence and development of ovary dysfunction induced by cisplatin and thus can provide diagnosis evidence for ovary dysfunction induced by chemotherapy.
