壳聚糖-β-磷酸三钙作为可注射组织工程骨支架材料的可行性研究
Feasibility of chitosan-beta-tricalcium phosphate as an injectable bone tissue engineering scaffold material
【摘要】 目的探讨以壳聚糖-β-磷酸三钙(β-TCP)作为骨髓基质干细胞(BMSCs)注射型骨组织工程支架材料的可行性。方法将体外培养并扩增的中国青山羊BMSCs与可注射的壳聚糖-β-TCP支架复合培养(材料组),以单纯接种培养的BMSCs为对照组,倒置显微镜观察细胞生长情况,MTT法检测材料对细胞增殖的影响。将BMSCs与壳聚糖-β-TCP混匀成BMSCs/壳聚糖-β-TCP复合物,扫描电镜观察BMSCs在材料表面的生长情况。结果倒置显微镜下见材料组细胞生长良好,与对照组无明显差异,MTT法显示材料组细胞增殖与对照组无显著性差异(P>0.05),扫描电镜下见材料表面细胞增殖良好。结论BMSCs/壳聚糖-β-TCP复合物表面细胞增殖良好,该生物材料具有良好的细胞相容性,可作为可注射型骨组织支架材料。
【Abstract】 Objective To study the feasibility of using chitosan-beta-tricalcium phosphate(beta-TCP)as an injectable scaffold material in vitro to deliver goat bone marrow stromal cells(BMSCs).Methods BMSCs from the bone marrow of a goat were cultured and induced,and the third passage of it was used to culture with beta-TCP,to compare with the cells cultured with pure fetal calf serum as the control group.The morphology and proliferation of the cells were observed under inverted phase contrast microscope.MTT assay was used to examine the effects of beta-TCP on BMSCs viability.BMSCs were mixed with beta-TCP to generate BMSCs/beta-TCP composite with final cellular numbers of 1×107.The morphology of the cells on the surface of beta-TCP was observed by scanning electronmicroscope(SEM).Results BMSCs co-cultured with beta-TCP exhibited good growth as observed under inverted phase contrast microscope,without significant difference from the control group.MTT assay showed no significant difference in the number of cells between beta-TCP and the control group(P>0.05).Under SEM,the BMSCs showed good adhesion to beta-TCP with obvious proliferation.Conclusion BMSCs can grow and proliferate well on the compound BMSCs/beta-TCP and beta-TCP has good biocompatibility with BMSCs in vitro,which may be used as a good scaffold material for injectable tissue engineering bone.
