高效液相色谱法测定头孢他啶血药浓度
Determination of Ceftazidime in Plasma by HPLC
【摘要】 目的建立测定头孢他啶血药浓度的方法。方法高效液相色谱法采用KromasilC18色谱柱(250mm×4.6mm,5μm),以甲醇-磷酸二氢钾溶液(15∶85)为流动相,流速1.0ml/min,检测波长254nm。结果头孢他啶血浆浓度在0.5~200.0mg·L-1范围内线性关系良好(r=0.9999)。低、中、高3个浓度的方法回收率为94.91%~105.95%,RSD<12%;提取回收率为89.50%~92.94%,RSD<5%。结论本方法简便、准确、精密度好,可用于临床药动学研究。
【Abstract】 Aim To establish a method for the determination of ceftazidime concentration in plasma. Method The HPLC separation was performed on Kromasil C18 column (250mm×4.6mm, 5μm). The mobile phase was methanol-potassium dihydrogen phosphate (15∶85) at a rate of 1.0ml/min. The detection wavelength was 254nm. Results The linear range was 0.5~200.0mg·L-1(r=0.999 9). The method recovery of ceftazidime was 94.91%~105.95% (RSDs lower than 12%) and the extraction recovery of ceftazidime was 89.50%~92.94% (RSDs lower than 5%). Conclusion The method appeared to be simple,accurate and precise. It is suitable for clinical pharmacokinetic study.
