【摘要】 目的观察加味左金丸对大鼠胃癌前病变环氧合酶-2(COX-2)蛋白表达的影响,并探讨其治疗机理。方法采用MNNG、0.03%雷尼替丁、56℃15%盐水、0.85%脱氧胆酸钠、40%乙醇、饥饱失常等多种因素制作大鼠胃癌前病变模型和免疫组化方法检测正常组、自然恢复组、加味左金丸高、中、低剂量组、维甲酸组大鼠胃粘膜组织中COX-2蛋白表达的情况。结果加味左金丸中、低剂量组COX-2蛋白染色积分与自然恢复组比较差异有统计学意义(P<0.01,P<0.05)),高剂量组和维甲酸组与自然恢复组比较差异无统计学意义(P>0.05);加味左金丸中剂量组COX-2蛋白染色积分与维甲酸组比较差异有统计学意义(P<0.01)。结论加味左金丸对COX-2蛋白的高表达有明显的抑制作用,其治疗胃癌前病变获效的机制之一可能与其在一定程度上通过调控多种癌基因、抑癌基因,降低COX-2蛋白的表达,进而恢复细胞增殖与凋亡的平衡有关。更多还原

【Abstract】 Objective: To observe the effect of adjusted Zuojin pills(AZP)on cyclooxygenase-2(COX-2) protein expression in rats with gastric precancerous lesions(GPL),and study the possible mechanism of AZP in treating GPL.Methods: The model was made mainly through MNNG together with other factors including hypso-concentration hot brine,sodium deoxycholate,alcohol stimulus,ranitidine,undue hunger,overeating.The expression of COX-2 protein was detected by immunohistochemical method in normal group,spontaneous recovery group,AZP groups of high-dosage,medium-dosage,low-dosage,retinoic(RA)group.Results: There were very significat difference between the medium-dosage AZP group and the spontaneous recovery group(P<0.01),and there were significant difference between the low-dosage AZP group and the spontaneous recovery group (P<0.05),and there were no significant difference between the high-dosage AZP group,the RA group and the spontaneous recovery group(P>0.05),and there were significant difference between the middle-dosage AZP group and the RA group (P<0.01),and there were no significant difference between the middle-dosage AZP group and the normal control group(P>0.05).Conclusion: AZP could treat rats with GPL by inhibiting cyclooxygenase-2 protein expression.更多还原
