用紫外分光光度法测定水相中甲壳素的含量
The Measurement of Chitin with UV Spectrophotometry
【摘要】 将甲壳素溶于NaOH/CO(NH)2水相溶剂中,研究了甲壳素水溶液的紫外光谱性质,确定甲壳素在新溶剂中的最佳紫外吸收波长为230 nm,试验了不同溶剂组分浓度、测试温度对紫外分光光度法测定甲壳素浓度的影响,发现对于不同溶剂组分浓度的甲壳素溶液,甲壳素的质量百分比浓度与紫外吸收值均有较好的线性相关性,且测定时温度在8-20℃范围内时,测定结果准确。
【Abstract】 Chitin was dissolved in NaOH/urea aqueous solvent and the UV spectrophotography plat of chitin was studied.The authors had an insightful investigation on the influence of different solvent composition and different measuring temperature for the measurement of chitin concentration by UV spectrophotometry.It was ascertained the maximum absorb-wavelength of UV was 230 nm,and there was favorable linear interrelations between the content of chitin and UV absorption with different solvent composition.Additionally,when the temperature is in the range from 8 to 20 ℃,the result of measure is very satisfying.
