甲基叔丁基醚对大鼠小脑急性毒作用研究
Acute Toxic Effect of Methyl Tertiary Butyl Ether in Cerebellum of Rat
【摘要】 目的观察甲基叔丁基醚(MTBE)对大鼠小脑皮质神经递质GABA、Na+,K+-ATP酶、Ca2+-ATP酶的影响,探讨甲基叔丁基醚对中枢神经系统的急性毒作用机制。方法54只健康成年SD大鼠,以染毒前为对照,染毒剂量为1050 mg/kg体重,染毒后用免疫组织化学方法检测大鼠小脑皮质神经递质GABA的表达并用相对灰度值定量;用分光光度法测定小脑皮质Na+,K+-ATP酶、Ca2+-ATP酶的活性。结果与染毒前比较,小脑皮质GABA在MTBE染毒后0.5 h时表达开始增加,1.0 h达到高峰,2.0 h时开始减弱,8.0 h时GABA表达恢复到染毒前水平;MTBE染毒后1.0 h小脑皮质Na+,K+-ATP酶、Ca2+-ATP酶的活性降低(P<0.05);染毒后8.0 h小脑皮质Na+,K+-ATP酶、Ca2+-ATP酶的活性无变化(P>0.05)。结论MTBE对小脑急性毒作用可能与神经递质GABA含量的改变有关;MTBE可降低大鼠小脑Na+,K+-ATP酶、Ca2+-ATP酶的活性,改变神经细胞能量代谢。
【Abstract】 Objective To explore the acute toxic mechanism of methyl tertiary butyl ether(MTBE),by detecting the change of the neurotransmitter γ-amino-butyric acid(GABA) and the activity of Na+,K+-ATPase,Ca2+-ATPase in the cerebellar cortex.Methods 54 adult SD rats were randomly divided into control and experiment groups.The MTBE exposed dose was 1 050 mg/kg.bw and respectively injected by abdominal cavity.The immunohistochemical method(SABC method) was used to detect the change of the neurotransmitter GABA in the cerebellar cortex.The spectrophotometric method was used to measure the activity change of the Na+,K+-ATPase,Ca2+-ATPase in the cerebellar cortex.Results The expression of the GABA in cerebellar cortex was changed in the different time(0.5、1.0、2.0、4.0、8.0 h) after exposed MTBE(1 050 mg/kg.bw).The expression of the GABA was began to increase in 0.5 h,reached peak in 1.0 h,began to reduce in 2.0 h,and was recovered to normal level in 8.0 h.The effect of MTBE on the activity of Na+,K+-ATPase,Ca2+-ATPase in cerebellar cortex was decreased dramatically in 1.0 h compared with control(P<0.05).The activity of Na+,K+-ATPase,Ca2+-ATPase was recovered to normal level(P>0.05)in 8.0 h. Conclusion(1)The acute toxic effect of MTBE in the cerebellum is relevant to the neurotransmitter GABA.(2)MTBE can reduce the activity of the Na+,K+-ATPase,Ca2+-ATPase,to affect the energy metabolism of the nerve cell.
