Bak和Bcl-xl在豚鼠形觉剥夺性近视眼视网膜的表达及凋亡

【摘要】 目的观察豚鼠形觉剥夺性近视视网膜变性中细胞凋亡,及Bak和Bcl-xl蛋白在视网膜细胞中的表达。方法将出生3天的豚鼠40只,单眼半透明眼罩遮盖,对则眼不做任何处理为对照。随机分为A组(遮盖8周)和B组(遮盖12周),分别于实验前及实验后行检影验光、A超测量眼轴,摘出眼球,用TUNEL技术检测视网膜细胞凋亡情况,用免疫组化法分析Bak和Bcl-xl蛋白表达水平。结果遮盖眼均较对照眼屈光度加深,眼轴延长,且随遮盖时间的延长而增长。TUNEL标记显示A组及B组遮盖眼视网膜内、外核层均发现凋亡细胞,两者凋亡率分别为(0.27±0.14)%和(2.33±0.37)%,差别有统计学意义(P<0.05)。遮盖眼Bak表达均较对照眼增高,且Bak在B组遮盖眼视网膜的表达较A组高,两者差别有统计学意义(P<0.05);Bcl-xl的表达在遮盖眼及对照眼之间无明显差别(P>0.05)。A组、B组遮盖眼视网膜细胞的Bak表达与视网膜细胞凋亡率的相关系数r值分别为0.79、0.84(P<0.05),与Bcl-xl的r值分别为-0.37、-0.31(P>0.05)。结论凋亡是豚鼠形觉剥夺性近视视网膜细胞变性的机制之一,Bak的高表达可能与形觉剥夺性近视视网膜细胞凋亡有关。

【Abstract】 Objective To observe the apoptosis and apoptosis related protein Bak and Bcl-xl expression on the retinal degeneration of form deprivation myopia (FDM) in guinea pigs. Methods 3 days old guinea pigs (n=40) were worn with monocular translucent goggles for 8 weeks(A group) and 12 weeks(B group) respectively. The contralateral eyes were used as control. Refractive state was monitored by retinoscopy and A-scan ultrasonography before and after experiment. Removed the eyeball, TUNEL method and immunohistochemical technique respectively for the detections of apoptosis and Bak and Bcl-xl. Results Ocular diopters and axial lengths in all occluded eyes remarkablly increased in comparison with its contralateral eyes. TUNEL-positive apoptotic cells were observed in retinal outer and inner nuclear layer of the occluded eyes, B group apoptotic rate was significantly more than A group [(0.27±0.14)%vs(2.33±0.37)%,P<0.05].Moreover,the expressions of retinal Bak protein of B group were higher than those of A group (P<0.05). The linear correlation coefficient between apoptosis and Bak expression of A group and B group were 0.79,0.84(P<0.05),and that between apoptosis and Bcl-xl expression were -0.37,-0.31 (P>0.05). Conclusion Apoptosis is one of the mechanisms of retinal degeneration of FDM, and the Bak expression may be associated with retinal apoptosis of FDM.
