RMB5A反义寡脱氧核苷酸对不同转移潜能大肠癌细胞系裸鼠移植癌体内影响研究
Inhibitory effect of RAB5A on the growth and metastasis of colorectal cancers in nude mice
【摘要】 目的观察RMB5A反义寡脱氧核苷酸对人大肠癌高、低转移潜能细胞系的裸鼠移植瘤的抑制作用及抑制肿瘤转移效果。方法建立人大肠癌高转移和低转移的裸鼠模型,分组给予RAB5A反义寡核苷酸治疗,观察荷瘤鼠的肿瘤体积、肿瘤大小以计算抑瘤率;RT-PCR和免疫组化法检测治疗组RAB5A的表达。结果在大肠癌裸鼠移植瘤高转移和低转移组中RAB5A反义寡核苷酸均具有抑瘤效果(P<0.05)。反义寡核苷酸组的肿瘤组织中RAB5A基因和蛋白质表达均减少,远隔脏器无转移发生。而正义寡核苷酸组以上指标均与模型组无差别(P>0.05)。结论体内抑制RAB5A可抑制裸鼠移植癌生长及推迟转移。
【Abstract】 Objective To observe the inhibitory effect of RAB5A on the growth and metastasis of colorectal cancers in nude mice.Methods Nude mice were inoculated with human colorectal cancer cells and cured with RAB5A sense and antisense oligonucleotides.The volumes and sizes of transplanted tumors were recorded and analyzed to obtain the inhibitory rates.RT-PCR and immunohistochemistry were used to detect the expressions of RAB5A in tumor tissues.Tumor metastasis in main organs were observed by pathological examination.Results RAB5A antisense oligonucleotides inhibit the growth of the high and low metastasis colorectal cancers(P<0.05).In the antisense oligonucleotide group,expression of RAB5A gene and protein reduced,no metastasis in other organs was observed,while in sense oligonucleotide group,these changes could not be observed(P>0.05).Conclusion The metastasis of colorectal cancer in mice could be suppressed when RAB5A expression is inhibited by antisense oligonucleotide.Antisensenucleotide RAB5A therapy could be an effective method for colorectal cancers.
