小鼠嗅感觉神经细胞的急性分离
A Method of Acute Isolation of Mouse Olfactory Receptor Neurons
【摘要】 目的建立一种急性分离小鼠嗅感觉神经细胞的方法。方法利用酶消化结合机械吹打方法,急性分离昆明小鼠嗅粘膜的嗅感觉神经细胞。结果本实验方法能分离出具有正常形态特征的嗅感觉神经细胞,细胞在体外可以存活5小时左右,可用于膜片钳等实验研究。结论本研究建立了一种快速、可靠、简便易行的急性分离成年小鼠嗅感觉神经细胞的方法,此法不损伤细胞膜的电生理学特性。
【Abstract】 Objective:A method is described for producing acute isolated olfactory receptor neurons from adult mouse nasal epithelium.Method:The olfactory receptor neurons could be isolated from olfactory mucous of Kunming mouse using enzymatic digestion and mechanical isolation.Results:Using the method of this experiment,the olfactory receptor neurons with normal configuration can be isolated from mouse nasal epithelium and the neurons can keep alive for 5 hours in vitro.These neurons can be used in patch clamp technique experiments. Conclusion:A acute credible simple and convenient method of acute isolation mouse olfactory receptor neurons was developed in this experiment.This method keeps the electrical characters of cell membrane.
