PKC和MMPs在枸杞籽油抗动脉粥样硬化中的作用和机制

【摘要】 目的研究枸杞籽油抗动脉粥样硬化的功效及其可能的作用机制。方法高脂膳食制备家兔动脉粥样硬化实验模型,观测其血浆中TC、TG、LDL-C、HDL-C、ApoA、ApoB的变化;56 d后,测定血清SOD、GsH-Px、T-AOC活性和MDA的含量;石蜡切片进行病理学检测;免疫印迹检测PKC、MMP-2、MMP-9在血管中的表达。结果枸杞籽油可增加实验家兔血浆中HDL-C、ApoA的含量,降低血浆中TC、TG、LDL-C、ApoB的含量,增强血清中SOD、GSH-PX、T-AOC的活性,降低血清中MDA的含量;降低PKC、MMP-2、MMP-9在血管中的表达。结论枸杞籽油具有显著的抗动脉粥样硬化效应。其机制可能是通过PKC途径,减少MMPs的分泌、抑制SMC的增生、分泌胶原、降血脂和抗氧化等。

【Abstract】 OBJECTIVE To study the anti-atherosclerosis potency of Lycium seed oil and its possible mechanism.METHODS The rabbit atherosclerosis model was established by high fat diet,and the TC,TG,LDL-C,HDL-C,ApoA,ApoB levels in plasma were examined dynamically.The SOD,GSH-PX,T-AOC activities and the MDA levels in serum were monitored after 8 week’s high fat diet.Aorta samples were observed for atherosclerotic extent,and the expression of PKC and MMPs were measured by immunohistochemistry.RESUITS Contents of HDL-C and ApoA obviously increased in plasma of low and high dosage Lycium seed oil groups,while TC,TG,LDL-C and ApoB levels significantly decreased compared with the control group.The SOD,GSH-PX,TAOC activities up-regulated while the MDA levels decreased in Lycium seed oil groups compared with the control group.The expression of PKC,MMP-2,MMP-9 were significantly lower than those in the control group.CONCLUSION Lycium seed oil has potent anti-atherosclerosis effects,its anti-atherosclerosis potency is similar with the lovastatin.The possible mechanism involves the decreasing of plasma lipids,anti-peroxidation and inhibiting the expression of PKC,MMP-2,MMP-9.
