白术多糖对离体蛙心生理功能的影响

【摘要】 采用不同浓度的白术多糖溶液和两种不同的受体阻断剂(β受体阻断剂-心得安和M受体阻断剂-阿托品)对蟾蜍进行离体心脏灌流,并对实验结果进行记录和统计分析。结果发现10μg/ml的白术多糖溶液即可加强心肌的收缩能力,二者呈现显著的量效关系。白术多糖溶液在一定浓度范围内不影响心率,但升高到500μ/ml时可使蟾蜍心率增加,至1000μg/ml时引起心肌过度收缩和心动过速,去药后出现心脏功能衰竭。在进行机理研究时选择100μg/ml的白术多糖浓度。结果表明:白术多糖对心肌的作用类似于肾上腺素,可被β受体阻断剂所抑制,M受体阻断剂对其作用几乎没有影响。讨论了心肌细胞和自律细胞对不同浓度白术多糖的敏感性以及可能存在的介导途径。

【Abstract】 Different concentration of polysaccharide from Atractylodes macrocephala and two kinds of receptor blockers(propranolol and atropine)were applied to isolated frog hearts perfusion and the results were recorded with the multi-channel physiological signal collecting system.The data show that the polysaccharide of 10μg/ml started to excite the heart contraction and the action increases with the increasing polysaccharide.The heart rate is not affected when the polysaccharide is within 10μg/ml to 100μg/ml but is significantly accelerated while it reaches 500μg/ml.The polysaccharide of 1000μgμg/ml results in excessive myocardial contraction and tachycardia,and heart failure when the drug is removed. The polysaccharide of 100μgμg/ml was applied in the related mechanism research.The results show that the effects of the polysaccharide on myocardium were similar to that of epinephrine,which could be blocked byβreceptor blockers,but not by M receptor blockers.The sensitivity of myocardium cell and pacemaker cell to the polysaccharide and the possible mediated pathway were discussed.
