【摘要】 目的:探讨益气活血方对心梗后大鼠缺血心肌局部血管新生的影响及其分子学机制。方法:建立心梗模型并分为5组(益气活血方大剂量组、益气活血方小剂量组、麝香保心丸组、假手术组、模型组),灌胃6周后把存活的大鼠处死,免疫组化法测定心梗边缘区新生血管数及微血管面密度;检测缺血心肌血小板源性生长因子-B(PDGF-B)蛋白的表达。结果:与假手术组相比,模型组大鼠心梗边缘区新生血管数、微血管面密度及PDGF-B蛋白表达明显升高(P<0.05);大剂量组和麝香保心丸组的新生血管数、微血管面密度及PDGF-B蛋白表达均有明显的升高,与模型组比较均有显著差异(P<0.05),小剂量组也有不同程度表达。PDGF-B蛋白表达与微血管数增多呈一致性。结论:益气活血方能够促进心梗后大鼠缺血心肌局部血管新生,对缺血心肌具有保护作用,上调心肌局部PDGF-B蛋白表达可能是其作用机制之一。更多还原

【Abstract】 Objective:To probe into the influence of Yiqi Huoxue Prescription on growth of ischemic myocardium vessel and protein expression of PDGF-B of rats after myocardial infarction and its molecular mechanism.Methods:The models of myocardial infarction were built and divided into five groups(large dosage group of Yiqi Huoxue Prescription,small dosage group of Yiqi Huoxue Prescription,Shexiang Baoxin Bolus group,sham operation group and model group).Six weeks after intragastric infusion,the survival rats were killed to detect its new-growth vessels’ numbers on the edge of myocardial infarction with the immunohistochemical method and the density of the microvasculature volume.The expression of platelet-derived growth factor hematoblasts B(PDGF-B)of ischemic myocardium was detected.Results:Compared with the sham operation group,rats’ new-growth vessels’ numbers on the edge of myocardial infarction,the density of the microvasculature volume and expression of PDGF-B in model group were apparently raised(P<0.05),and so were those of large dosage group and Shexiang Baoxin Bolus group.There were also expression to different degree in small dosage group.The expression of PDGF-B and the number of the microvasculature appeared homogeneous increase.Results:Yiqi Huoxue Prescription can promote the growth of ischemic myocardium vessel of rats after myocardial infarction and protect ischemic myocardium.And enhancing the expression of PDGF-B in myocardium may be one of its mechanisms.更多还原
