重组人Ⅱ型肿瘤坏死因子受体-抗体融合蛋白对慢性阻塞性肺疾病大鼠营养状态的影响
Effects of Recombinant Human Tumor Necrosis Factor-Fc on Nutritional Status of Rat Model of Chronic Obstructive Pulmonary Disease
【摘要】 目的探讨重组人Ⅱ型肿瘤坏死因子受体-抗体融合蛋白(rhTNFR:Fc)对慢性阻塞性肺疾病(COPD)大鼠营养状态的影响。方法48只大鼠随机分为4组:正常对照组、COPD组、rhTNFR:Fc干预组和干预对照组。单纯吸烟法建立COPD模型,后3组烟雾暴露达1个月时,干预组给予rhTNFR:Fc进行干预,干预对照组给予空白对照制剂。熏烟80d称重后,处理所有大鼠。正常对照组、COPD组肺组织切片行HE染色观察形态学改变,定量测定肺平均内衬间隔(MLI)、平均肺泡数(MAN)。以酶联免疫吸附法(ELISA)测定各组血清中的TNF-α浓度。使用自动生化分析仪测定各组大鼠血清白蛋白含量。结果①COPD组肺MLI高于正常对照组,MAN低于正常对照组(P<0.05)。②COPD组血清TNF-α浓度高于正常对照组和rhTNFR:Fc干预组(P<0.05),和干预对照组比较差异无显著性(P>0.05)。③COPD组体重较正常对照组、rhTNFR:Fc干预组体重降低,差异有显著性统计学意义(P<0.01)。rhTNFR:Fc干预组体重与干预对照组体重比较,差异有显著性统计学意义(P<0.01)。④COPD组血清白蛋白较正常对照组降低(P<0.05),但与rhTNFR:Fc干预组和干预对照组比较差异无显著性统计学意义(P>0.05)。⑤(COPD组+干预对照组)大鼠血清TNF-α浓度与体重呈负相关(r=-0.75,P<0.05),与血清白蛋白浓度呈负相关(r=-0.73,P<0.05)。结论rhTNFR:Fc可降低COPD大鼠血清TNF-α水平;rhTNFR:Fc可能对改善其营养状态具有一定作用。
【Abstract】 [Objective]To investigate the effects of recombinant human tumor necrosis factor-Fc (rhTNFR:Fc)on nutritional status of rat models of chronic obstructive pulmonary disease (COPD).[Methods]Forty eight rats were randomly divided into a normal control group, COPD group, rhTNFR:Fc-interfered group, sham interfered group.Rat models of COPD were established by exposure to cigarette smoking daily.After 1 month , rats in interfered group were treated with rhTNFR:Fc, sham interfered group injected with certain preparatum as control. On the day 80, in normal group and COPD group,lung tissure section stained by hematoxylin and eosin(HE)was observed to study the morphological alterations .Mean linear intercept(MLI)and mean alveolar numbers (MAN) were also measured. The levels of TNF-α in serum were detected with ELISA , levels of albumin in serum measured by automatic biochemistry analyzer.[Results]MLI was higher in COPD group than in the normal control group, while MAN were on the contrary. The levels of TNF-α in serum of COPD group were significantly higher compared with normal control group (P<0.05 ) and rhTNFR:Fc-interfered group (P<0.05 ). The weight of COPD group was significantly decreased compared with the normal control group (P<0.01 ) and rhTNFR:Fc-interfered group (P<0.01 ).No differences were found between the weight of COPD group and the interfered control group(P>0.05 ),significant differences found between the weight of rhTNFR:Fc-interfered group and interfered control group(P<0.01).The levels of albumin in serum of COPD group were significantly decreased compared with normal control group(P<0.05),but no differences were found among the levels of COPD group and rhTNFR:Fc-interfered group and interfered control group(P>0.05). The levels of TNF-α in serum of COPD group and interfered control group revealed negative correlations to the weight (r = -0.75,P<0.05 ) and the levels of serum albumin (r = -0.73,P<0.05).[Conclusion]rhTNFR:Fc can reduce the levels of TNF-α in serum of rat model of COPD .It may play certain role in the improvement of nutritional status of rat model of COPD.
