托吡酯对戊四氮点燃大鼠海马结构白细胞介素-6表达的影响
Effects of topiramate on the expression of IL-6 in the hippocampus of pentylenetetrazol kindling rats
【摘要】 目的:探讨戊四氮(PTZ)点燃大鼠海马结构白细胞介素-6(IL-6)表达的特征及托吡酯(TPM)对其表达的影响。方法:将30只成年健康雄性SD大鼠随机分为3组,每组10只。Ⅰ组大鼠灌胃生理盐水1h后,腹腔注射10g/LPTZ生理盐水溶液(35mg/kg);Ⅱ组大鼠按40mg/kg灌胃10g/LTPM生理盐水溶液1h后,腹腔注射PTZ(剂量同Ⅰ组);Ⅲ组大鼠按10ml/kg和3.5ml/kg分别胃灌注和腹腔注射生理盐水。各组每d给药1次。当Ⅰ组大鼠癎性发作行为达到点燃标准时,观察各组大鼠的行为,记录脑电图(EEG),采用免疫组织化学SABC法检测海马结构IL-6的表达。结果:给药4周,Ⅰ组大鼠点燃成功,而Ⅱ组大鼠最高发作级别较I组低,对照组大鼠行为正常。Ⅰ、Ⅱ组大鼠海马结构IL-6表达较Ⅲ组增加(P<0.05),且Ⅰ组的表达高于Ⅱ组(P<0.05)。结论:癫癎大鼠免疫系统处于活化状态;TPM能降低PTZ点燃大鼠海马结构IL-6的表达而对癫癎具有一定的免疫抑制作用。
【Abstract】 Aim: To explore the effects of topiramate (TPM) on the expression of interleukin-6 (IL-6) in the hippocampus of pentylenetetrazol(PTZ) kindling rats.Methods: A total of 30 healthy male rats were randomly equally divided into three groups.The rats of group Ⅰ were given normal saline 4 ml/kg by gavage 1 h, then intraperitoneally injected with 10 g/L PTZ normal saline daily. The rats of groupⅡ were given 10 g/L TPM normal saline 40 mg/kg by gavage 1 h, then injected with PTZ normal saline daily. The rats of group Ⅲwere given the same amount of normal saline as other groups daily. When the rats in group Ⅰ were kindled successfully, the behavior and EEG of the 3 groups were observed, and the IL-6 level in the hippocampus of all rats were detectd using immunohistochemistric SABC method. Results: The rats in group Ⅰ reached kindling criterion after 4 weeks, while the spasm scale of the rats in groupⅡ was lower. The IL-6 levels in the hippocampus of group Ⅰ and groupⅡ significantly increased compared with group Ⅲ (P<0.01). Furthermore, the IL-6 expression of group I was higher than that of groupⅡ (P<0.05). Conclusion: The immune system of the rat epileptic models is greatly activated, and TPM can decrease the expression of IL-6 in the hippocampus of rats kindled by PTZ,which might provide some protective action through immunosuppression.
