慢性阻塞性肺疾病大鼠气道炎症细胞及黏蛋白检测
Detection of airway inflammation and Mucin5AC in rats with chronic obstructive pulmonary disease
【摘要】 目的:制备大鼠慢性阻塞性肺疾病(COPD)模型,对支气管肺泡灌洗液(BALF)进行细胞学检查,并观察黏蛋白(Mucin5AC)的表达。方法:20只Wistar大鼠随机均分为健康对照组和COPD组。以气管内注射药物和吸烟方法制备COPD模型,6周后收集BALF进行细胞学检查,以免疫组化方法观察呼吸道杯状细胞肥大情况及Mu-cin5AC的表达。结果:①BALF检查:健康对照组中性粒细胞数为(17.58±3.72)×106L-1,COPD组为(251.10±42.76)×106L-1,2组差异有统计学意义。②气道炎症病理学评分:健康对照组为(18.56±2.46)分,COPD组为(69.30±4.99)分,2组差异有统计学意义。③肺泡腔直径:健康对照组为(47.22±12.95)μm,COPD组为(92.58±4.32)μm,2组差异有统计学意义。④杯状细胞化生指数:健康对照组为(5.12±3.07)个/mm2,COPD组为(51.20±9.23)个/mm2,2组差异有统计学意义;⑤Mucin5AC阳性染色面积:健康对照组为(3.12±2.60)%,COPD组为(34.50±6.52)%,差异有统计学意义。BALF中的中性粒细胞计数与气道炎症病理学评分呈正相关。结论:多种炎症细胞参与了COPD发生及发展。
【Abstract】 Aim: To detect the airway inflammation and Mucin5AC in rats with chronic obstructive pulmonary disease (COPD).Methods:Twenty Wistar rats were divided randomly into 2 groups: normal group (group N, n=10) and the model of COPD group (group C, n=10). The COPD model was established through passive smoking and intratracheal instillation of LPS. BALF was collected to count inflammatory cells after 6 weeks. Immunohistochemistry was processed to observe the goblet cell and the expression of Mucin5AC in airway tissue of the rats.Results: BALF analysis presented that PMN count was higher in group C ((251.10±42.76)×10~6 L~ -1 ) than that in group N (( 17.58 ±3.72)×10~6 L~ -1 ) and there was significant difference; airway pathological score was higher in group C (69.30±4.99) than that in group N ( 18.56 ±2.46) and there was significant difference; alveolar diameter was longer in group C ((92.58±4.32)μm) than that in group N ((47.22±12.95)μm) and there was significant difference. DAB staining presented that goblet cell hyperplasia index in group C ((51.20±9.23)/mm~2) was higher than that in group N ((5.12±3.07)/mm~2) and there was significant difference; Mucin5AC was overexpressed in group C ((34.50± 6.52 )%) and almost absent in group N (( 3.12 ±2.60)%) and there was significant difference. The PMN count was correlated with the airway pathological score.Conclusion: The results suggest that many inflammatory cells involve in the development and progress of COPD.
