天麻钩藤饮对脑缺血大鼠的神经保护机制研究

【摘要】 目的观察天麻钩藤饮对脑缺血大鼠学习记忆能力,VEGF及其受体VEGFR-2/FLK-1的影响,探讨天麻钩藤饮的脑保护作用机制。方法采用大鼠双侧颈动脉永久性结扎制备脑缺血模型,于术后次日开始给予天麻钩藤饮〔5g/(kg·d)〕灌胃,分别于2周及4周通过Morris水迷宫观察大鼠行为学的改变,免疫组化观察VEGF及FLK-1的改变。结果模型组大鼠的潜伏期较假手术组明显延长,记忆能力明显下降。治疗组较模型组明显好转,4周较2周更为明显。2周模型组VEGF、FLK-1主要在神经元表达,较假手术组明显增多,4周表达下降;治疗组VEGF、FLK-1在神经元的表达较模型组明显增多,4周仍高表达。结论该研究提示,天麻钩藤饮可能通过促进VEGF和特异性受体FLK-1的表达,启动神经元保护机制,发挥脑保护作用,改善脑缺血后大鼠的行为学表现。

【Abstract】 Objective Using a cerebral hypoperfusion model,we investigated the effect of Tianmagouteng Yin on cognitive dysfunction in rats,and investigated the expression of vascular endothelial growth factor(VEGF) and its receptor VEGFR-2/FLK-1 in order to clarify its mechanism of actions.Methods Cerebral hypoperfusion was induced by permanent occlusion of both the common carotid arteries(2VO).Then the next day of insult rats were administered Tianmagouteng Yin(5g/kg·d).After 2 or 4 weeks of treatment,the performance in Morris water maze was investigated.The presence of VEGF and its receptor FLK-1 were investigated by immunohistochemistry.Results After 2 weeks of treatment,2VO rats administered Tianmagouteng Yin showed significantly improved latency in Morris water maze compared to model control groups,and after 4 weeks of treatment,it improved even more.The number of VEGF and FLK-1 positive cells increased significantly after treatment.And the positive VEGF and FLK-1 cells were mainly neurons.The expression of positive cells was still in high level at 4 weeks.Conclusion These results indicate that Tianmagouteng Yin has an ameliorating effect on the spatial memory deficit caused by cerebral hypoperfusion,and it is suggested that Tianmagouteng Yin may exerts a direct action on neurons that partly mediated by VEGF/FLK-1 pathway.
