【摘要】 目的建立甲氨蝶呤血药浓度测定方法。方法采用高效液相色谱法。色谱柱:C18反相柱(200 nm×4.6 nm,5μm),流动相:甲醇∶磷酸缓冲液(pH 7.2)为17∶83,流速:1.0 mL/m in,柱温35℃,检测器波长306 nm。结果线性范围0.25~100.0μg/mL,线性关系良好,回收率101.74%,精密度RSD<5%。结论该方法简单快捷,灵敏度高,结果准确,适用于甲氨蝶呤血药浓度监测。

【Abstract】 Objective To establish an HPLC method to assay of methotrexate(MTX) in plasma in order to monitor its plasma etmoentrafion in vivo.Methods The concentration of MTX in the sample was determined by RP-HPLC,with a YWG C18(200 nm×4.6 nm,5 μm)column and at the temperature of 35 ℃.The mobile phase was methanol-0.01% phosphate solution(17∶83) with a flow rate of 1.0 mL/min.The detected wavelength was set at 306 nm.Results Good linearity was shown over the concentration range of 0.25-100.0 μg/mL.The average recovery was 101.74%,The relative standard deviation of within-day and between-day were less than 5%.Conclusions This method is concise,convenient,high sensitive and accurate to monitor the concentration of MTX in plasma.
