TDI在小鼠血清中的代谢及代谢产物的鉴定
Study on metabolism and metabolite of TDI in mice serum
【摘要】 研究甲苯二异氰酸酯在小鼠血清中的代谢情况.采用高效液相色谱法,色谱柱:ODS(250 mm×4.6 mm,5μm);流动相∶乙腈∶水(45∶55)(加0.22 g醋酸钠).在1.2~5.2 mol/L范围内,甲苯二异氰酸酯峰面积与浓度线性关系良好(r=0.999 8),甲苯二异氰酸酯在雄性小鼠血清中代谢生物半衰期为51.72 min,甲苯二异氰酸酯在雌性小鼠血清中代谢生物半衰期为111.77 min.用气相色谱-质谱法测定甲苯二异氰酸酯(TDI)在小鼠血清中的代谢产物,结果显示,2,4-TDI被代谢成2,4-TDA.
【Abstract】 To test the metabolism of TDI in serum.High Performance Liquid Chromatography was applied to determine the residual concentration of TDI after different time incubation in serum.C18 column:ODS(250 mm×4.6 mm,5 μm);mobile phase:acetonitrile:water(0.22g ammonium acetate).In the range of 1.2~5.2 mol/L,there existed a good linear correlation for toluene diisocyanate(r=0.999 8),the half life of TDI in male serum was 51.72 min,and 111.77 min in female.Metabolite of toluene diisocyanate(TDI) in mice serum were studied by Gas Chromatography-Mass Spectrometric(GC-MS).The result reveal 2,4-TDI has metabolized and the metabolite is 2,4-TDA.
