高效硫氧化菌的筛选及其应用
Isolation and application of high efficient sulfur-oxidizing bacteria
【摘要】 从含有硫化氢气体的制药废水中利用平板划线法分离出两株化能自养硫杆菌zw1与zw2.考察zw1与zw2的形态特征与生理生化特性以及接种了zw1与zw2的生物滴滤塔对污水处理厂水解池产生的硫化氢气体的去除能力.结果表明:zw1为短杆,好氧菌,革兰氏阳性,适宜温度为22~32℃,适宜pH为5.0~7.0.zw2为长杆,兼性好氧,革兰氏阴性,适宜温度为22~25℃,适宜pH为4.0~6.5.接种了zw1与zw2的生物滴滤塔具有较强的去除能力与抗冲击负荷能力,容积负荷≤192 g/(m3.h)时,去除率接近100%.zw1与zw2的代谢产物以硫酸根为主,不会造成滴滤塔的堵塞,可长期稳定运行,易于实现工业化.
【Abstract】 Two autotrophic rod sulfur-oxidizing strains (zw1 and zw2) were isolated from Pharmaceutical wastewater containing hydrogen sulfide gas,and were streaked and purified by repeated transfer to fresh solid medium on the plate.Shape,physiological and biochemical characteristic of chemolithoautroph zw1 and zw2 was checked,and removal capacity of hydrogen sulfide gas from hydrolyzation tank in wastewater treatment plant was investigated by the inoculated biofilter with zw1 and zw2.The results suggested that the optimal temperature of zw1(aerobic bacilli,short rod and Gram-positive) was between 22 ℃ and 32 ℃ and the optimal pH was between 5.0 and 7.0.And the optimal temperature of zw2(facultatively-aerobic bacilli,long rod and Gram-negative) was between 22 ℃ and 25 ℃ and the optimal pH was between 4.0 and 6.5.Furthermore,the inoculated biofilter with zw1 and zw2 had stronger ability to resist shock of high inlet loading.Removal efficiency of hydrogen sulfide was close to 100% when the maximum volumetric loading achieved 192 g/(m3·h).Metabolism products of zw1 and zw2 were mainly composed of SO42-and the biofilter was not blocked during the experiment of more than three months.Therefore,the biofilter need not carry out a back washing frequently and it can be operated steadily for long-term.It is a potential industrialization method that can be fulfilled easily.
