RhoA抑制剂C3转移酶对大鼠肝星状细胞黏附和迁移的影响
Effects of RhoA inhibitor C3 transferase on the HSC adhesion and migration
【摘要】 目的观察RhoA抑制剂C3转移酶对肝星状细胞(hepatic stellate cell,HSC)增殖、黏附和迁移的影响。方法将培养的肝星状细胞分为4组:对照组,LPA组,C3组,LPA+C3组。采用甲苯胺蓝染色法测定细胞黏附率,Boyden Chamber小室测定HSC跨膜迁移数量,用四甲基偶氮唑盐(MTT)法检测细胞增殖。结果C3转移酶对HSC的增殖、黏附和迁移均具有抑制作用,并且抑制了LPA诱导的HSC增殖、黏附和迁移。结论RhoA信号转导通路抑制剂-C3转移酶对LPA诱导的HSC增殖、黏附和迁移具有抑制作用。
【Abstract】 Objective To observe the effects of RhoA inhibitor-C3 transferase on the proliferation,adhesion and migration of hepatic stellate cell(HSC).Methods The cultured HSCs were divided into 4 groups,which were control group,LPA group,C3 group and LPA+C3 group.The HSCs adhesive rate was detected by toluidine blue colorimetc assay,the counts of migration of C3 transferase of HSCs were tested by modified Boyden chamber,and cell proliferation was determined by MTT assay.Results RhoA inhibitor-C3 transferase inhibited HSC proliferation,adhesion and migration,and inhibited the LPA-induced HSC proliferation,adhesion and migration.Conclusion RhoA inhibitor-C3 transferase could inhibit LPA-in duced HSC proliferation,adhesion and migration.
