亚慢性吸入二硫化碳对小鼠免疫细胞损伤研究
Study on Damage of Immunological Cells Induced by Sub-chronic Inhalation Carbon Disulfide in Mice
【摘要】 目的:研究二硫化碳对小鼠免疫细胞的损伤。方法:以浓度为0,400,800,1200 mg/m3的二硫化碳蒸气给小鼠做静式吸入染毒5周,每天2 h。用单细胞凝胶电泳实验和流式细胞术检测二硫化碳对白细胞的损伤。结果:低、中、高染毒组与对照组小鼠白细胞彗星拖尾率经2χ检验,差异有统计学意义(P<0.01);各试验组与对照组彗星头部百分比差异有统计学意义,并且各个试验组之间差异也有统计学意义(P<0.01);各试验组与对照组尾长差异亦有统计学意义,并且各个染毒组之间差异存在统计学意义(P<0.01)。试验组与对照组生殖细胞的凋亡率分别为13.50%,22.10%,33.60%和0.2%,各剂量组与对照组之间差异有统计学意义(P<0.01)。结论:亚慢性吸入二硫化碳可损伤小鼠白细胞DNA并促进白细胞调亡。
【Abstract】 Objective: To study the damage effect of inhalation carbon disulfide on immunological cells of mice.Methods: The damage of white blood cells of mice exposed to carbon disulfide by two hours static inhalation per day at different concentrations(0,400,800 and 1 600 mg/m3) for five weeks was analyzed with single cell gel electrophoresis(SCGE) and flow cytometry(FCM).Results: There was a significant difference between the experiment groups and the control group(P<(0.01)) in the rate of cells with tail;and there was also significant differences among the experiment groups and the control group in the percentage of head of the comet and in the length of tail(all P<(0.01).) The rate of apoptosis in all groups were 13.50%,22.10%,33.60% and 0.2% respectively,with significant difference among the experiment groups and control group(P<(0.01).) Conclusion: The sub-chronic carbon disulfide exposed can induce DNA damage and the apoptosis of white blood cells.
