DNA损伤信号作用下的肿瘤抑制因子的反馈环模型
A Feedback Loop Model of Tumor Suppress Gene Network by Activation Responding to DNA Damage
【摘要】 P53肿瘤抑制因子在基因调控网络中起重要作用.作为对外界刺激的响应,P53可以激活或调节下游基因的表达.最新的实验结果表明在单细胞中存在P53和MDM2的持续振荡.本文讨论P53和MDM2蛋白系统的负反馈环模型.在适当引入DNA损伤信号条件下,可以描述P53和MDM2系统的调控特性以及典型的振荡行为.
【Abstract】 P53 tumor suppressor plays a key role as a factor in gene regulatory networks. In response to cell stress, P53 is activated and regulate the transcription of its downstream genes. Recent experimental results revealed sustained Oscillation levels of both P53 and Mdm2 in individual cells. In this paper, we discuss a negative feedback loop model to study the dynamicalmechanism ofP53and Mdm2proteinsystem. Thetypicaloscillatory behaviors forthe activities ofP53 and Mdm2 proteins in individual cells are described in a way through properly introducing the dynamics of DNA damage signal.
